Two other Orbitraps run with Proxeon ion sources connected to other manufacturers LCs, but we see a lot of carryover when compared to EASY-nLC. We don't really need blanks between samples. Ran the EASY-nLC system for 2 weeks straight before changing the buffers and used only 10mls buffer A and 5mls buffer B!!!!!”

“We decided on a split-less system for several reasons:
1) Better retention time reproducibility: with a split you run a "constant pressure" system with flow rate changing if the pressure changes due to sample conditions (high protein level, impurities, etc.). The split-less systems are "constant-flow rate" systems which will keep the same flow rate despite any pressure changes due to sample conditions. This is more important to us on our triple quad where we want to use scheduled SRM experiments, but also very useful in our label-free quantitation experiments on our OrbiTraps. 2) Easier to maintain: the Proxeon system has much more user serviceable parts, many being simple chromatography parts you already have lying around. 3) Solvent use: now we can buy small quantities of more expensive solvents which result in better instrument signals. We also have only about 1L of organic waste to dispose of every 3 months. 4) Easier to teach grad students and post-docs if you do not have to get them to make and maintain consistent splits. It saves me time as well. You also have fewer chromatography plumbing connections that may leak. 5) We have made extensive use of the ability to use the autosampler pump as a third system pump to run 2D online chromatography separations (Journal of Proteome Research, ACS, DOI: 10.1021, 2009). 6) The small size allows us to position the unit at right angles to the source of all our Thermo instruments so the overall dead volume in the system is reduced, saving us time on each chromatographic run.”
